Of National scope.

Caloric restriction is a metabolic intervention that has been shown to extend the lifespan and healthspan in a wide range of animals, including mammals. The fact that primates show positive effects in response to intermittent fasting protocols provides a rationale for testing its impact on human ageing. Of special clinical interest are the the strategies to treat the ageing phenotypes of post-menopausal women, due to the clinical relevance of this cohort and the lack of useful treatments However, we currently lack specific molecular biomarkers that can measure amelioration of ageing in organs and could thus be used to assess the success of such interventions. We recently described and validated a series of specific markers of senescent cells. Since accumulation of senescent cells in tissues has been linked to ageing, we propose to use these novel markers to determine for the first time the impact of intermittent fasting on postmenopausal women ageing. Our objectives are to (1) validate ours and other markers in the context of caloric restriction, and (2) use these markers to determine organ ageing in post-menopausal women undergoing intermittent fasting. To reach these goals, we will use a combination of in vivo models, biochemical analyses, proteomic/metabolomic screens and dietary interventions in human volunteers. The final goal is to propose easy and inexpensive protocols to ameliorate ageing in this cohort.
